I bought 54 box's of the SK-FRESCA tile and was wondering if I could put a good wet look Glaze on them.

I'm not familiar with those--do you have a link t the manufacturer?

Will these be used for a floor?

My guess is no---if they are glazed , anything you use to make it shiny or wet looking will wear off of the glazed surface----just a guess without knowing the tile----
