I'm planning to install 2 new light switches. In the attached picture is it acceptable to run 12/3 from the right junction box to the left junction box to power switch 2? Or do I need to run two distinct 12/2 wires between the junction boxes?

Yes that is acceptable.
The only thing I would change is to use the red as the switched conductor between the switches and the black as the constant hot. Personal choice, not required.
