Walls or floor?
Typically a chalk line is used to snap a straight line down the center of the room----then a measurement is made of the tile --plus one grout line-(actually two tiles and two grout lines-) and that measurement is used to make a line near the wall---
If the second line is 1/2 of a tile or larger--you are good in that direction---
If it is less than 1/2 of a tile---move the center line over by 1/2 tile and use that line to measure over towards the wall----
Once that line is complete---repeat the process in the other direction---

I have also found that you want to check what the tile configuration will be around the toilet. You don't want to have to cut thin slivers of tile, or a three-quarters inner crescent around the flange. As Mike suggests above, get the two perpendicular lines in the "center" of the bathroom, but check edges and around toilet flange, vanity, any other built-in to make sure you don't have a tiny piece at the edge. Move the "center" point over to increase the small piece or eliminate it completely.

What kind of tile are you using?
Like the guys above said, you don't want to end up having little slivers of tile on the sides.
If your tile is not square, you want to make sure you pick the right 'direction'. If the tiles are rectangular, you want them to go in the long direction.
And if you do have one of those really odd shaped areas, think about turning the tiles 45 deg. While it's a bit more tile cutting....it can solve odd shaped area layouts. If you get our line right...the pieces you cut in half get used on each side...with maybe a little trimming.
