You will need to remove the caulking. What type did you use and where? When applying the cauling again, tape off both edges and tool the caulking with a metal spoon dipped in denatured alcohol to give a perfect smooth joint.

When we caulk we always use a wet rag to remove any edges. Its difficult to remove these edges once they have been painted over. You can try a very sharp scraper. Remove the caulk and sand area smooth. Just remember after you caulk an open joint, smooth with finger, then wipe with a wet rag removing excess caulk. Good luck.
