I have a suspended three car garage floor (approx 750 sq ft). I want to frame out a couple of rooms under the floor. If the walls are anchored to the floor and walls, is it necessary to anchor to the suspended floor above? I am not going to frame in the ceiling (bottom side of the garaage floor) and would prefer not to anchor or disrupt the integrity of the garage floor.

Yes. you need to secure the floor to the ceiling. What is the floor structure of this suspended garage floor?
Post some photos from below and above.

In looking at my question, it may not be too plain as to what I'm trying to do. The area that I want to frame out is under the suspended floor. The top plate would butt up against the suspended floor. I need to know if anchoring the top plate to the suspended floor would damage the integrity and warranty of the suspended floor or if even if it is necessary given that the base plate are anchored to the basement floor and walls.

Warranty questions would be answered by the builder.
I was just curious as to the consrtuction of a suspended garage. That was the reason for the picture request.

We are out of state right now. Otherwise, I would post a picture (assumming I have the tech know how to do it), but it is an amazing thing. The entire area is supported by the outside walls and it is rated to 200K pounds. Still scares me though. My plan is to contact the contractor for final word, just curious if anyone knew anything about them.
