guys - i'm building a shower, with one of those glass frameless doors. How do I make sure it has enough support to open/close without falling off? A standard piece of 2"x4", nailed in like a stud where the hinges are expected to be? How about a piece of 4"x4" instead - greater strength and a bit more flexibility on hinge placement?

Any advice on this would be greatly appreciated!

Those doors are heavy, so there needs to at least be a stud where the hinges land. A double stud(jack/king) as in a standard door opening is ideal.

which is stronger 2 2x4, or 1 4x4?

2-2x4 glued together. In effect, it is a lam beam.
