See my attached drawing. While you are pretty much correct on the vertical pipe load- as soon as it turns horizontal under the floor you have to reduce the load to 8 DFU's. You are maxed out with the 3 fixtures shown. In fact, you might be over loaded depending on Cal sizes. You need to dump that into a 3" bldg. drain

oops, you mention turning into a 3" Would you turn into 3" where the two 2" meet on the first floor and then bring 3" into the ground and to the main sewage line? Or bring both 2" pipes through into the ground and connect at a wye into a 3" and then run that 3" (roughly 15ft) to the main 4" drain?

The horizontal lines needs to be 3" based on your load.
Personally, i'd put a 3x2x2 y above the floor.
