Insulating Wooden Shed/Outbuilding

Hello everyone! Im going to be insulating my 10x16 high barn wooden shed. The shed has the lofts on both sides for storage. I have a couple of questions. The shed come with an eve vent on both sides of the shed near the peak. Will I need to cover those up or leave them open for ventilation? Also, the ceiling does have a radiant barrier installed under the wood and shingles as you can plainly see and read that's what it is. Should I insulate between the 2x4 studs of the ceiling as well? I plan on installing a window AC unit for the summer and possibly space heaters for the winter. Thanks in advance for any help/advice!
