Plywood, tile board, toilet flange, spacer?

Doing a bathroom remodel...

I have a toilet flange screwed directly into a plywood sub floor.

I want to tile over the plywood but want to put down a 1/4 in tile board first. If I do that, and then put penny tile sheets down, will I need a spacer over the toilet flange or will the height created by the tile board and the tiles themselves be small enough that no spacer would be needed?

Any access from below to the plumbing?
If so consider cutting the tail piece under the flange and removing the whole thing flange and all, install the flooring then drop the tail and flange in place and reconnect using a Fernco.
Not willing to do that then a double sized wax ring will do the job.
