Vent slope question

1. Does the vent (dry) require a 1/4" slope just like any other drain line?

2. I need to run the vent line in the first floor's joists. They are 10x2 spaced 16". Is it safe to drill 2 1/2" holes through 6 consecutive joists?

If I need a 1/4" slope, I figure I could drill the first 2 1/2" hole at 3" inches from the top of the joist, then this would put the last hole would at 5" from the top.
