Feed boiler off pex manifold or main supply ?

Going to start redoing my house with a pex manifold.

Would it be better to have a cold water feed off my pex manifold that goes to my boiler or have it "T" off my main 3/4 supply line before it goes into the manifold?

I realize both will work, but what is typically done in this case?

thanks

Is the manifold going to have shout off valves to each of the pex runs?

Are you using a mana block system?

I would run off the manifold.
