Double Vanity Spacing & Rough Drain Position

I'm struggling with deciding how to space the sinks & medicine cabinets for a new bathroom double vanity. The space is 77.5 inches wide with stud walls on both sides. I'm roughing in the drains and I'm pretty sure they need to aligned perfectly with the sinks...

Should I simply divide 77.5 into thirds, and center each sink/drain/medicine cabinet at 25.83" from each end? The two medicine cabinets are 20" wide (and will be centered above each sink), so that would leave 12.5" from the medicine cabinets-wall, and 12.5" between medicine cabinets.

Would it be better to increase the gap in the middle? Opinions???

(I will probably end up having to custom build the vanity and counter top - I've never done this before )

You should probably go by standard cabinet sizes, not by the space you have. If something ever needed to be swapped out, nobody is going to want to have a custom made vanity put in there again.

I've found that although 6' is a lot for a single vanity, it's not quite enough for two. It's also awkward to have the vanity cabinet right next to a side wall, you tend to bump your elbows a bit.

your spacing would work fine. what about using two 36 inch cabinets with a 3 inch filler at each end ripped to fit the space? your sink center would end up about 21 inches from each end and you could use standard vanity cabinets.
