12' is a stretch for a traditional multiple 2x header. you should be looking at lvl, steel beams or similar.
since this is the second floor, need to consider bearings below, likely down to grade. to echo what bob22 said, this isn't something to guess at. if looking at lvl, lots of lumber yards will size the beams for you if you give them some plans of what you are trying to do. most do it for free or free if you buy the lvl from them.
when you start talking middle floors of multi-story buildings, however, you may be headed for the consultation of a structural engineer.
