framing opening

Have a 2"x 4" 16" on center exterior wall 8' high double top plate and single bottom plate. On top of the wall are the ceiling joists and roof rafters for the attic. The 2"x 4" wall sits on a 1"x 4" floor board. The flooring in the rest of the area has been replaced with 3/4" plywood. The exterior wall sits on top of 2"x 8" flooring joists which is on a 2"x 4" wall. Want to frame an approx 36" wide opening 5' high. The opening will not go to the outside for a door but for a recess in the wall. Do the vertical studs to support the ends of the header need to line up with the 2"x 8" floor joists or can they be on top of the 1"x 4" floor board. Thanks.

The studs can sit on top of the floor but do they need to be in line with the 2" x 8" joists. Thanks.
