Measuring Supply and Return FPM

I've completed my crawlspace HVAC upgrade from R4.2 flex to R8 pipe with dampers. Attic is next.

What is the best way to measure air flow FPM? I have a handheld anemometer. Is the best method to remove the deflector grills and place the unit flush with the opening and centered. I have noticed that the air flow measures higher or lower in different locations.

Are you tying to determine CFM from through the supplies and returns. if so, then the covers must be on, and an average reading across the face is used.
