Cold Air return
I have a question.
A central A/C and heat pump has been put in my home, there are 2 bedrooms, 1 bath, and an open floor plan that joins the livingroom and kitchen.
There is 2x12 supply vents in the house- 1 in each bedroom, and 2 in the living room... will be adding more eventually. These are all 6 inch flex ducting

The cold air return is wide open to the basement and currently is drawing air through the basement door- I need to add a cold air return vent-- do I add 1 in the central part of the house in the floor near the thermostat- or would it work to us 6 inch flex ducting and a 2x12 in each room as a return? It has to be in the floor because the walls are plaster and are too skinny to accommodate ducting.

Open air returns in a basement, finished or not, are a bad idea overall. While a central, ducted return from a side wall or a ceiling would be better, it still isn't ideal.

What is best is to have individual returns in each room if that is possible. It will insure that, even with the door closed to the room, you will get the right amount of air flow to properly heat/cool the room. Central returns need to be able to pull the air back to the air handler from each room. If it cannot, it can cause uneven heating/cooling in rooms.

Your overall duct sizing for returns is another matter entirely. If the duct isn't sized properly you may not be bringing enough air back through the unit and that can cause coil freezups, poor heating and premature failure of components because of the added work the unit must do to compensate. This is one of those times where you may want to think about getting a professional to look at the system. If that is not an option for you then we may be able to help you but it is going to require more information overall.

Put a large return near the thermostat, and 1 in each bedroom(7" flex to a 4x10 in bedrooms).
