what size and how many vents are required for a room 64'x 40'

If my duct calculations are correct (most likely not and I'll soOn be corrected) and a 12" duct can carry 700 cfm than 3 12" ducts @ 700 cfm and one 8" duct which would carry 350 cfm to accomodate your need for 2560 cfm for the area of that room.

You'll need to add a return to pull that air out as well for free flow or it can lead to static pressure issues, restriction on the air flow felt by the entire system leading to poor heat extraction due to poor evaporation, high(er) energy costs, etc.

Doc, I probably left out too many things. I was asked to finish the ac installation that some else started. I do residenual installations and small commercial ones. They have 2 5 ton condensors and air handlers and 12" and 8" flex duct work. The air handlers are going to be mounted on a stand in the room and ductboard will go into the ceiling and branch off with the flex. How many supply air grills will I need and what size
