Closing fireplace - Air brick Question

I live in a 2 floor house + a basement. The fireplace is on the 1st floor and there is what appears to be a cleanout in the basement.

I want to close it up, so my plan is to brick off the hole and drywall over it.

My question(s) are :

1) I'm going to glue the drywall to the brick. Can I drywall over an air brick or is that too tight of an air gap?

2) Can I eliminate the air brick all together and leave the cleanout open in the basement?

Thanks!

Ok so what's "air brick"?
Cap off the top and any openings.
Install strapping to the brick with Tap Con screws, then sheetrock.

In the UK you have to ventilate an unused flue because of damp problems.
