do we need a sill plate under the door? Can the slider sit flush to floor opening?

Modern doors have a high threshold, 1+" to accommodate carpet. Can you set the door flush with the floor, yes, but it may be a lot of work. Setting the door on the subfloor as suggested, gets it 3/4" down with hardwood floors.

Why are you trying to get rid of the threshold?
Thresholds give definition between rooms and help with bridging the transition between flooring
